산수유 추출물이 Hydrogen Peroxide로 손상된 배양 NIH3T3 섬유모세포에 대한 세포붿�착등 및 산화적 손상에 미치는 영향 (Effect of Cornis Fructus Extract on the Cell Adhesion Ability and Oxidative Stress in Cultured NIH3T3 Fibroblasts Injured by Hydrogen Peroxide)

To clarify the effect of Cornus fructus (CF) extract on the cultured NIH3T3 fibroblasts injured by hydrogen
peroxide (H2O2), this study was carried out 새 examine the protective effect of cell adhesion ability and
oxidative stress of H2O2 in cultured NIH3T3 fibroblasts. For this study, XTT assay for the cell adhesion ability,
lactate dehydrogenase (LDH) activity and suproxide dismutase (SOD)-like activity for oxidative stress of H2O2
were done in the cultured NIH3T3 fibroblasts, after NIH3T3 fibroblasts were incubated with various
concentrations of H2O2 or CF extract. In this study, H2O2 decreased the cell adhesion ability of NIH3T3
fibroblasts in dose- and time-dependent manners. And XTT50 value was determined at 25uM of H2O2 which
was highly toxic by the criteria of cytotoxicity. In the effect of CF exreact on the H2O2-induced cytotoxicity, CF
extract significantly increased cell adhesion ability injured by the cytotoxicity of H2O2 in cultured NIH3T3
fibroblasts. CF extract also deceased LDH activity which was increased by the H2O2-induced cytotoxicity, and
showed SOD-like activity. From above the results, it is suggested that H2O2 was highly toxic on cultured
NIH3T3 fibroblasts, and also, CF extract showed the protective effect on oxidative stress of H2O2 by the
showing of SOD-like activity and the decrease of LDH activity.
